Source:
Antiserum (Rabbit)
Immunogen:
Human LONG®R³IGF-I
State and Appearance:
Lyophilized powder
Specificity:
As determined by RIA: Antiserum is specific for LONG®R³IGF-I
No significant cross-reactivity with human, rat or chicken IGF-I
No significant cross-reactivity with human IGF-II, human insulin or porcine growth hormone.
Storage/Stability:
At least 6 months at 2 - 4°C (lyophilized)
After reconstitution, aliquot and store at -20°C
Avoid freeze-thaw cycles
Reconstitution:
Dissolve the vial contents in 250 µl RIA buffer (30 mM sodium phosphate pH 7.5 containing 10 mM EDTA, 0.2 g/l protamine sulphate, 0.5 ml Tween 20, 0.25 g/l sodium azide) to give an antiserum stock of 1/50.
Application and Titre:
Radioimmunoassay 1/20,000
At a dilution of 1/20,000 the antibody binds 25 - 30 % of fresh [125I]-labelled LONG®R³IGF-I tracer (80 Ci/g).
Half-maximal inhibition (ED50) of radioligand binding is produced by 200 - 250 pg of LONG®R³IGF-I and the limit of detection is 10 pg/tube.
Protocol:
#3002: Determination of IGF-I or IGF-II in a Range of Species by Radioimmunoassay (RIA)
