Source:
Antiserum (Rabbit)
Immunogen:
Recombinant rat IGF-I
State and Appearance:
Lyophilized powder
Specificity:
As determined by RIA, using PAT2:
100 % rat and mouse IGF-I
<0.05 % human IGF-I, chicken IGF-I
0 % rat IGF-II, human IGF-II
Storage/Stability:
At least 2 years at 2 - 4°C (lyophilized)
After reconstitution, aliquot and store at -20°C
Avoid freeze-thaw cycles
Reconstitution:
PAT2 (for RIA): Dissolve the vial contents in 250 µl RIA buffer (30 mM sodium phosphate pH 7.5 containing 10 mM EDTA, 0.2 g/l protamine sulphate, 0.5 ml Tween 20, 0.25 g/l sodium azide) to give an antiserum stock of 1/50.
Application and Titre:
Radioimmunoassay 1/60,000
At a dilution of 1/60,000, the antiserum binds 25% of fresh [125I]rat IGF-I tracer.
Half-maximal inhibition (ED50) of radioligand binding is produced by 0.2 ng of rat IGF-I.
For Western Immunoblotting use anti-rat IGF-I (PAT1). Dilution 1/1,000
For IHC, use IGF-I Antibody for immunohistochemistry. Dilution 1/50 - 1/200
Protocol:
#3002: Determination of IGF-I or IGF-II in a Range of Species by Radioimmunoassay (RIA)
